Purification of the Major Soybean Leaf Acid Phosphatase That Is Increased by Seed-Pod Removal.

چکیده
Fruit removal for 5 weeks after flowering increased acid phosphatase activity 10-fold in soybean (Glycine max L. Merr. Var Hobbit) leaves compared with normal seed-pod-bearing plants. The major acid phosphatase activity in leaves was purified over 2700-fold, yielding a single polypeptide of 51 kD with a specific activity of 1353 units/mg protein using p-nitrophenylphosphate as the substrate. Isoelectric focusing demonstrated that the purified protein co-migrated with a majority of the activity that increased in leaves following seed-pod removal. Immunoblot analysis demonstrated that at least part of the increased activity was due to an increased abundance of the phosphatase protein. In situ enzyme activity staining localized most of the total phosphatase activity to vascular tissues, the leaf paraveinal mesophyll cell layer, and the lower epidermis. This distribution and the response to seed-pod removal paralleled previous results for soybean vegetative storage protein (VSP) [alpha] and [beta]. However, in a native polyacrylamide gel the VSP detected by immunological staining of electrophoretically transferred protein did not migrate with the majority of the phosphatase activity. Fractionation of crude leaf protein on concanavalin A-Sepharose yielded a fraction containing 97% of the total VSP but only 0.1% of the total acid phosphatase activity.

Improving soybean leaf physiological responses under drought stress by increasing seed zinc and iron concentration
To study the effects of both drought stress and seed zinc and iron concentrations on the photosynthesis, chlorophyll fluorescence, and proline and carbohydrate accumulations in soybean (Glycine max (L.) Merr. cv. M9) leaf, a split plot experiment was carried out in 2012 based on a randomized complete block design with three replications in greenhouse conditions at Yasouj University. Drought str...
متن کاملEffects of some PGRs on seedling emergence and CAT and POD activity of maize under low temperature stress. Hamid Reza Eisvand*, Nasim Fathi and Darioush Goudarzi
Low soil temperature is one of the reasons for poor germination and establishment of maize. The aim of this study was to evaluatethe possibilityof improvingthe seedling emergence and performance of maize under low temperature stress. A pot experiment was conducted on Zea maize (single cross 704) at 14 ºC as cold stress and seed treatments were priming with 200, 300 and 400ppm of GA3; 100, 200...

Timing, intensity, and frequency of biomass removal PREDICTING YIELD LOSS IN affects soybean yield. Yield reduction is less sensitive INDETERMINATE SOYBEAN FROM to biomass removal during vegetative growth because soybean can develop new leaf area that can compensate POD DENSITY USING SIMULATED for temporarily reduced assimilatory capacity. Singer DAMAGE STUDIES (2001) reported that yield reduct...
متن کاملEffect of water stress during seed development on morphometric characteristics and dormancy of wild radish (Raphanus raphanistrum L.) seeds
The effects of water stress on morphometric characteristics of wild radish seeds as well as the expression of its seed dormancy were investigated in a pot trial at Roseworthy, South Australia in 2005. This experiment was carried out in a completely randomised design with five water regimes (control, S4, S5, S6 and S8) and four replicates. The water stress was exerted by withholding irrigation a...
